Cryptococcus species are known to cause opportunistic infection, mainly in the lung and CNS in immunocompromised patients.

This is an USG guided abdominal lymph node biopsy from an HIV positive patient.

C. neoformans is known to infect in immunocompromised individuals, whereas C. gattii infects immunocompetent individuals

Variably sized (Round/ovoid 5-15 micrometres μm in diameter). Thick mucopolysaccharide capsule with refractile centre.

Narrow based “Tear drop-shapped" budding pattern (useful to differentiate from Blastomyces and Histoplasma).

Antiphagocytic and poorly immunogenic sugar capsule composed mainly of glucuronoxylomannan (GXM), which is a major virulence factor.

H and E staining: Cryptococcus yeasts have a clear capsule surrounding the pale blue nucleus.

Special stains:

PAS and GMS stains help demonstrate the yeasts.

Mucicarmine stain highlights the highly characteristic gelatinous capsule a bright pink (useful to differentiate from Blastomyces).
